Rickettsia IFA
Rickettsia are intracellular Proteobacteria
associated with arthropods, including fleas, body lice, mites
and ticks. The antibody response to infections by these agents
is primarily directed against a lipopolysaccharide (rLPS) and
1-2 outer-membrane protein (rOmpA and/or rOmpB) antigens. The
standard IFA and MIF methods detect antibody against the whole
organism and remain the most commonly used of the recommended
methods. Due to the range of antigens found on the whole
organism, however, this broad reactivity must be compensated for
by increasing the cutoff titer to 1:64-1:128. Generally, these
methods discriminate between the typhus group and spotted fever
group, although titers often differ between the specific
infecting species and related species by 2-4-fold. Our MIF
slides now offer reference laboratories the ability to make
these discriminations at the species level.

Rickettsia ELISA
ELISA techniques have
ranged from whole cell lysates to
individual purified antigens, with a
range of effects. Our lysate-type
antigens have been replaced by two
current formats. The rLPS-coated plates
often show the earliest antibody
response and they also demonstrate the
broadest reactivity within the typhus or
spotted fever groups. IgM
cross-reactivity to other bacteria
limits this assay to IgG antibody
testing (screening).
The best ELISA data
is now seen with the use of the
immunodominant outer membrane protein
(rOmpB). These proteins are
affinity-purified and show excellent
group sensitivity, with added
sensitivity for the particular species
the antigens were isolated from.
